The Jamaican world record holder had to come from behind to take the win in a photo-finish with his fellow countryman and training partner Kemar Bailey-Cole, who clocked 10.09 and Antigua’s Daniel Bailey, who posted 10.23 for third.
American Mark Jelks ran 10.28 for fourth place.
Bolt was hoping to get closer to the world-leading 9.86 seconds posted by USA’s Tyson Gay at the Jamaica Invitational on Saturday, but fell well short of that mark tonight.
“It wasn’t the best start for the season,” a disappointed Bolt said after the race. “I didn’t felt as great as felt in training.
“It was just a bad race. Got to go back to the drawing board and work on a few things and just get it right the next time.”
Carmelita Jeter dominated the women’s race to win in a world-leading time of 10.95.
The reigning World champion who skipped the Jamaica Invitational last week to recovery fully from beat Barbara Pierre of USA who ran 11.02 and Jamaica’s Kerron Stewart, who posted a time of 11.10.
Elsewhere at the meet, Novlene Williams-Mills of Jamaica won the women’s 400m dash in 50.73, beating Moushaumi Robinson of USA and she clocked 51.19.
The men’s race went to Luguelin Santos of the Dominican Republic in 45.45, with Jarrin Solomon (TNT) 45.65 and Riker Hylton (JAM) 45.95, completing the top three.
American Lashawn Merritt took the men’s 200m in 20.26, while Jamaica’s Hansle Parchment women the men’s 110mH in 13.25, equalling the world-leading mark. He beat Bahamians Ryan Brathwaite 13.26 and Shane Brathwaite 13.44.
