Half-centuries from Darren Bravo (82) and Denesh Ramdin (57) helped Trinidad and Tobago to 226 all out off 50 overs, but Barbados eased to 227 for seven off 48 overs to win the game.
For the Red Force, Rayad Emrit took three wickets for 45 runs in the defeat.
Guyana Jaguars booked its place in the semifinals after finishing second in Group B behind the Windward Islands.
In their last outing, the final round of the Regional Super50 at Warner Park on Sunday, the Jaguars defeated Combined Campuses and Colleges Marooners by five wickets.
In that game, Guyana knocked off the 255 target set for victory after they reached 255 for five off 49.3 overs with Assad Fudadin slamming an unbeaten 103 and Leon Johnson making 56.
CCC had made 254 for seven off 50 overs with Rovman Powell making an undefeated 63 and Anthony Alleyne scoring a half century of 57.
With the ball for the Jaguars, Raymon Reifer and Gudakesh Motie had each taken two wickets.
Experienced top-order batsman Shivnarine Chanderpaul has played 408 games for Guyana.
West Indies batsmen Darren Bravo and Narsingh Deonarine, and wicketkeeper Denesh Ramdin have all played over 100 games for Trinidad and Tobago.
