Luck, who had started every single game of his NFL career, was forced to sit out the last two games with an injured right shoulder.
However, after taking part in full practice throughout the week, the 26-year-old has been upgraded from questionable to active and will start at Lucas Oil Stadium.
He will take over from veteran Matt Hasselbeck, who led the team to two successive wins.
Luck has passed for 753 yards and thrown five touchdowns passes from three games this season.
At the same time though, the former Stanford star has also thrown seven interceptions, which had many fantasy owners pulling their hair for three successive weeks.
Colts (3-2) have won its last three games.
Meanwhile, Indianapolis is also listing running back Ahmad Bradshaw and linebacker Jerrell Freeman – who only recently signed – active for Sunday night’s contest.
Running back Dion Lewis, who was questionable with a “minor abdomen injury” is also active for Patriots.
Lewis was seen going through his regular pre-game warmup and test, but will wait until kickoff before deciding if he will play.
Tom Brady and the Patriots head into this contest as 9.5 point favourites, and hold a six-game winning streak against the Colts.
This is the first meeting between these sides since the “Deflategate” dispute following the AFC championship game which New England won.
