Klopp replaces the sacked Brendan Rodgers, following the 1-1 draw with Everton prior to the international break.
The team news sees Divock Origi selected to lead the attacking line for the Reds, with the Belgian supported by Philippe Coutinho and Adam Lallana.
Striker Daniel Sturridge was not in the matchday squad for Liverpool
Emre Can, Lucas and James Milner all start as well.
Klopp is without striker Danny Ings and defender Joe Gomez, who are both out for the rest of the season with a knee ligament and ACL injuries, respectively.
The club revealed the setbacks earlier this week and the pair now join captain Jordan Henderson (foot), striker Christian Benteke (hamstring) and Roberto Firmino (back) on the sidelines, although the latter trio are expected to return at some point during the season.
Meanwhile, for Spurs, Mousa Dembele starts alongside Dele Alli in midfield.
Tottenham manager Mauricio Pochettino is without three central midfielders, as Ryan Mason (knee) and Nabil Bentaleb (ankle) are both injured and Eric Dier is suspended.
Summer transfer window recruit Son Heung-Min is also out with a foot injury.
Spurs are sitting eight in the EPL standings with 13 points, while Liverpool are in 10th place on 12 points heading into Week 9.
“Liverpool are coming with a different manager, a different style, different ideas but the most important thing is how we take the game,” Pochettino said.
“We need to be focused on our job and try to deliver. We need to start the game very aggressively and be very focused on our job with no special attention on how they want to play.”
